Try to purify your agrobacterium culture before switching to Pcr. you take your cultivation of agrobacterium, made a series of perpendicular streaks with a handle, after 24 hours you take a single colony to redo the same thing until you get a pure culture. check after by gram staining and microscopic observation.
Yes that was good suggestion from Prof. Yuan. However you can also not also overlook the suggestion of Prof. Hayat, for better and reliable result. Purifying your bacterial culture is very much important in fact the success of transformation totally depend on the purity of your bacterial culture.
If you know the characteristics of the plant tissue well after the transformation, it is possible to do the test suggested by Dr Yau, otherwise it may be an extra expense.
In any case, the purification step mentioned above by Dr Essalmani is very easy and effective for purification of your strain and should solve your problem.
Could you attached a picture for us. I don't know what you really meant about "particles'. I used to grow LBA4404 in full strength of LB medium, I saw some 'white stringy things' in the culture after overnight. By reducing the salt (NaCl) of LB to half, the situation improved. I am wondering whether you had the same situation.
