I asm currently working with alginate and silk fibroin hydrogels. I prepared a blend of alginate and aqueous silk fibroin (straight from dialysis) and resorted to ionic crosslinking to form firm gels. After 7 and 14 days of culturing (ECs and/or fibroblasts), I found globular domains of SF on the surface of the hydrogels. At the same time, a lot fewer cells a a lower metabolic activity were found on these gels, comparatively to alginate-only. I related this to the conformational change of SF from random coil to beta sheet, that has been proven to kill cells (https://www.nature.com/articles/srep31037 ). However, this was only tested for encapsulated cells. Since mine were cultured at the surface of the gels, I am not confident that this is a suitable explication. I tried to find articles relating to silk fibroin conformational change and cells toxicity/down regulation of cell adhesion, however, I could not find anything. Has anyone heard of a similar phenomenon?
