I have a problem with cultivation of bovine turbinate cells on 48 well plate. Approx. 1 hour after medium change I have cytotoxicity in some wells on the side of the plate. Do you have any idea what´s happened to my cells?
Thank you very much Sneha Hande for your answer! the coulour of the medium in the these wells is the same as in the healthy ones, so I think the pH is ok.
It could be a special problem with the cell-type, but I have a very trivial query.
Are you regular user of multiwell format or u use it only for these cells?. Just because some cells are very sensitive to drying out. If u are not used to the multiwell format and if u take long time to change medium, then some initial well will show some toxicity effects (cells looking like skeleton). Just have a look whether u always see this in the wells, kept without medium for longer before looking for more complicated solutions.
okay.. this may not be accurate answer but.. do you think cells are not adhering? if they don't adhere .. after media change.. they probably are washing out (am assuming when u say toxicity they are actually dying)?. So may be coat one of the column/plate and try or keep without changing media for couple of hours and try!
Btw sorry that I don't have any idea about what are turbinate cells!
Do you have try another kind of plate? Do you coat the wells with gelatine or something different? Sorry, I don't know turbinate cells but I had a similar "problem" with primary cells and the problem was in aspiration of the gelatine after coating.
