I want to change a Cys for an Ala in my plasmid and I want to use a kit, there are many companies that offer this type of kits, which kit would you recommend me? I have never done this before so I want to use something simple and effective.
I have tried many kits, some based on PCR, others on Kunkel-type procedures using single-stranded DNA, etc. but in terms of efficiency and simplicity nothing beats Agilent's Quikchange kits (formerly from Stratagene). And if short of money, you can homebrew your own kit with Pfu polymerase and Dpn I.
QuickChange Site Directed Mutagenesis Kit, Stratagene (Now sold by Agilent) is excellent.
Phusion Site-Directed Mutagenesis Kit from Thermo Scientific is good one.
