I want to functionalise thiolated oligonucleotides with gold nanoparticles.
To this I need to adjust the ph of the solution to 3, add Na+, mix, then bring the solution back to 7.
I cant be sure till i get accurate measures of resuspended lyophilised DNA concentrations but my solution will be somewhere between ~ 500-1000 micro litres.
Im probably only going to need to do this procedure once. Is there any chance I could use litmus paper rather than buy a micro electrode pH sensor? Its a heck of an investment for a 1 use item.
I afraid for its accuracy. Litmus paper often indicates the range. I do not think you are able to determine the exact value.
You need a pH meter. I did not realize that the entire University of Huddersfield does not possess a pH meter despite the success of the soccer team at present. Probably the cost of a pH meter is minor in relation to the experimental and thesis value of your gold nanoparticles.
Universal indicator papers will slightly be more accurate than litmus paper, at least it gives a wider range of values. Those should be cheaper than a pH meter
thanks for your answers. the university does possess pH meters: Just not one small enough to measure my sample. when my success matches that of Huddersfield town FC I will surely be able to order my own.
