Hi

I have problems with my PCR negative controls, Sometimes I see a band in the gel electrophoresis in these controls (same master mix that in samples but without DNA template). The band is shorter than the target amplicon but it appears between 50 and 75bp. This kind of band appears sometimes in the samples too: I get the target amplicon size band(145bp) and the other band.

I have checked my reagents, cleaned my place of work, pipettes, changed tubes...in order to eliminate possible contamination but I still see these bands in the negative controls.

Could I have primer dimers of this size (50-75bp) even though my primers are around 20 bp?
