I isolated the ovarian cancer cells(Mainly with cubic morphology) from ascite fluid of patients, these cells were positive for epithelial and ovarian cancer markers such as E-cadherin,EPCAM,Cytokeratin7 and 18 ,...even poor expression mesenchymal markers such as vimentin was also seen,so I checked expression of mesothelium markers such as calretinin and podoplanin in these cells and they were positive.in this test I used from epithelial ovarian cancer cell line skov-3 as negative control(I found from articles that skov-3 was negative for mesothelial markers)but skov-3 was positive for mesothelial markers, can anyone help me How can I be sure of the nature of my cells? Thank you for your attention.
