Dear Ali, yes the concentration of the parasite DNA will be very low but there is no way how to extract only parasite DNA and other human DNA present in the blood sample to leave on the filter paper.

do you have any option how to measure the concentarion of DNA in your sample? You can find out if any of DNA is in your sample.

Maybe you could decrease the volume of dilution buffer in the final step of DNA extraction to increase the concentration. Be careful, Qiagen do not recommend less then 100 ul (but you can use 50 ul two consecutive times) It is because you have to cover the whole filter in the spin column by dilution buffer.

There is also possibility to concentrate extracted DNA by using of 100% ethanol - then centrifugation, drying of pellet (10 minutes)  and dilute again in the smaller amount of dilution buffer (best over night).

DNA can be extracted from two Ø 3 mm punches (≈ 6–10 μL blood) of the DBS on filter papers using the Chelex®-100 extraction method [18,19] with some modifications. In brief, two punches were incubated in 0.5% Saponin in PBS for five min. and washed in PBS for 30 min.at room temperature. The samples were then incubated for ten minutes at 95°C in 100 μL of 10% Chelex-100 (Bio-Rad Laboratory, USA). After centrifugation, 45 μL of the DNA eluate was transferred into 96-well plates and stored at -20°C until use.
