I am looking at analytes in sputum supernatants via ELISA (MSD). When sputum is processed in our lab, 8x the weight of the plug in PBS is added to the plug before dispersion and then centrifugation and a supernatant is produced. I am aware that other labs add different volumes of PBS (e.g., x3) to the plug at the initial stage. Some labs appear to correct for the diluent effect on analyte corrections where as others do not.

Should I correct my analyte concentrations (pg/ml) for the initial dilution (e.g. x8) +/- should I adjust my reported LLOD?
