Hello,
I have read in a few publications that to extract total DNA from sputum samples, both bead beating and enzymes are used to disrupt the cells. Bead beating is supposed to be efficient in disrupting "hard to lyse" cells.
Any one can comment on the advantage of combing these two techniques?

Dear Meriem, uniform mechanical lysis of all microbes is achieved by bead beating and lysis enzymes (lysozyme and lysostaphin) also help degradation of bacterial cell walls. Action of Lysostaphin is digestion of peptidoglycan of staphylococci in the samples. The usage of these two enzymes helps to extract all bacterial DNA from sputum samples especially for NGS and metagenomic studies.
