After Sputum decontamination and digestion with NALC (for MDR TB and XDR TB test) I store the decontaminated sputum in PBS. After two weeks of storage at 2-8 degrees celsius, I run the two tests side by side: rpoB,katG,inhA genes tested side by side with gyrA, gyrB, rrs, eis genes test. Why is it, I get good results for rpoB,katG,inhA genes test BUT NOT for gyrA, gyrB, rrs, eis genes test?

Does the PBS solution affect the stability of gyrA, gyrB, rrs, eis genes test FASTER compared to stability of rpoB,katG,inhA genes?

What is the stability of rpoB,katG,inhA genes COMPARED to gyrA, gyrB, rrs, eis genes?
