HLA-elution protocols for discerning immunopeptidomes are getting increasingly used in recent publications on auto-immune disorders, in search of potential epitopes.
Most utilize blood samples, but frequently these diseases are focal. Blood samples supply abundant material for HLA-elution discovery, but samples from i.e. synovial fluid, sputum, cerebrospinal fluid may have very low cell numbers.
I was able to find a publication describing a protocol capable of identifying HLA bound peptides from as few as 10 million cells / mL. Does anyone know of sensitive protocols capable of detecting peptides from less cells?
