For vius recovery, Kindly introduce more details regarding your samples' type, and what  you are intending  to do exactly. Also, do you want to make absolute quantification?

Hello, In a first sight, I can see you don´t have enough virus in your samples to reach the concentration AND  the volume you want. I mean, if you have 200 mL and want 50 mL you could only concentrate your sample 4 times, so, a sample of 1x10^3 would be 4x10^3. If you want to increase your viral concentration to 10^6 you will have to concentrate your sample 1000 times. So I think It is kind of impossible. Otherwise, if the volume is not a problem for you, I can recommend using Centrifugal Units (Merk Millipore, Amicon), so you can get rid of the solution and concentrate your virus easily. Thank you for your reply. My sample is the virus laysate. Usually, after CPE, I freez-thow  my culture flask 4 times. then centrifuge at 3000 rpm for 10 minutes. Then I get the cDNA and run qPCR to estimate the virus concentration in the sample as plaque assay does not work with me. My virus is rotavirus A. and yes, I compare the concentration of my virus to the standard curve.

I want to have a better concentration, so is there any way to concentrate the virus laysate that I have now at 10^3 with out lossing a huge volume. what is the best method.

In my opinion, since you have cell lysate, you can go through a second passage which will increase your viral titer. Also, you can use PEG method for viral concentration but it will concentrate other proteins
