I am working on a tissue culture project on bulbous monocot plants, which twin scales are my ex plants for the callus induction. All the callus i got from different treatments are compact callus. Is there a way to convert compact callus to friable callus please?
Dear Nuwan Sameera Liyanage I never heard before about this possibility. Likely, a compact callus might be used as starting materials to obtain friable (morphogenic) calli. However, I can't see any sense on it. Better use suitable explants for calli induction from the beginning. The suitable fate (shoot or embryos regneration) for calli can be ruined easily. In my experience, estrict steps must befollowed, otherwise could be obtained nothing.

Dear Nuwan Sameera Liyanage as Ricardo Julian Licea-Moreno I have never heard on this possibility. I also worked in callus induction on some Amaryllidaceae species (bulbous monocots as well) and they easily develop callus from leaf-base explants (at early stages of leaf development). Also, very young fruits, and flower buds are commonly used to induce morphogenic calluses in this botanical family. Depending on your project goals, I would recommend different things (based in my experience). If you wish to regenerate plants from callus; then, flower buds, young fruits, and leaf bases are oK (however the culture conditions to regenerate them can be very different to those employed to grow calluses). If you wish to stablish a cell suspension culture (e.g. to study the biosynthesis of specialised metabolites) these calluses are not appropriate as often you can't obtain a real cell suspension culture, and metabolic profile (also yield of biochemicals) are reported to be low in undiferenciated cell clumps or tissues obtained from these species. In any case, I would not use twin explants (or any type of bulb explants) for callus induction as (in my experience) they develop more abnormal bulblets rather than calluses. As mentioned by other scientists, the choice of proper explant is a pre-request to have a successful in vitro regeneration. In addition, choosing an appropriate culture medium (MS, B5, MT, etc.), vitamin mixture, and hormonal balance play key roles in successful regeneration. I've experienced indirect regeneration using TCL explants in the fig tree.

Dear Nuan. In some monocots the combination of different auxins and immature flower buds may work better. For example in some banana varieties, a combination of IAA (1 mg/L), NAA (1 mg/L), 2,4-D (4 mg/L), L-Glutamin (100 mg/L) and Biotin (1 mg/L) are required to get friable embryogenic callus, from immature male flower bus. The gelling agent and its concentration need also be considered. Once you put the explants in the media and culture it in the dark, you have to wait 4-5 months to get the embryogenic callus. No change of media during this time. In some other experiments authors reported changing the media (same as above but 2,4-D 1 mg/L) after 1-2 months and obtained friable embryogenic callus. There is also an article that talk about induction of callus with Fexepid (FPX) in short, in some dicots and monocots. Try different ways and not repeat the same thing if you don't see some promising results.... All the best!
