Hi,
O’Brien et al. suggested using Toluidine Blue O dye to polychromatic staining of plant cell walls in 1964. Staining by O Toluidine blue provides reddish purple in collenchyma and parenchyma, red in Sieve tubes and companion cells, reddish purple or red in un-lignified compound middle lamellae. Cotton blue and Safranin O double-staining make a clear distinction between the fungal cell walls (blue) and the plant cell walls (orange) in epidermis, collenchyma and parenchyma tissue (Marques et al. 2013). It seems the red color you are discussing about may be related to Safranin O.
