I am evaluating RNA integrity from Leishmania using the Agilent 2100 Bioanalyser, but I frequently obtain low RIN values (~4-6) even when the RNA appears intact on an agarose gel. Since Leishmania naturally cleaves its 28S rRNA, I suspect this affects the automated RIN calculation. Has anyone else encountered this issue? What would be a more suitable alternative for assessing RNA quality in trypanosomatids?
