I have cellular culture growing at 28°C (teleost fish), and have continuously preserved that temperature during centrifugation and suspension. I marked a counter of that and put 200000 in a eppendorf tube and put fluorochrome (FITC-IgM) indierctly; this Ab need around 4°C incubation. I think that cells would become streesed or simply die around 10% of total. What do you think about the viability?
Storage of cells at 4°C is routine in cellular protocols. for example before flow cytometry and immunofluorescence methods ( as you mentioned) for prevention of unspecific antibody binding, long distance cell transfer, etc. So, don't worry but be careful about details of protocol.

many protocols for flowcytometry on human cells require incubation at 4 degrees. But you are working with another species. So you may heve to experiment your self. and if certain percentage of your cells die how would this affect your experiment?
