Given the cost and delay of plasticware, I'm wondering how many have had success coating their own plates? Specifically, I'm interested in protocols for primary trout hepatocytes on collagen plates, and the total concentration of collagen per well you've used. There's plenty of older protocols available, (circa 80s, 90s), but does anyone do this one a regular basis? If so, are there any specific details you'd share for success?
Hello Jason - no specific help for your specific cell type, but I have nearly always made my own collagen-coated plates for human HPCs and endothelial cells (sticking with concentrations from the older c1990s papers - which for endos were revised a bit from the pre-90s papers). It does matter if your collagen is resuspended thoroughly and stored properly, but other than that was/is straightforward.
