Is anyone familiar with pattern of CD80/CD86 expression on monocytes before and after stimulation with various stimuli e.g. LPS, IFNg, LPS/IFNgamma
In my experience, CD80 was highly expressed on monocytes, while CD86 expression was lower.
In addition, stimulation of monocytes with LPS alone seems to result in reduced expression of CD80/86, exactly the opposite as with dendritic cells. Does anyone have experience with monocyte activation?

I can offer some insight on part of your question, CD86. In my research, CD86 was a cell surface marker for monocyte/macrophage/myeloid lineage, characterizing the cells that I was using as such (THP-1, U937, HL-60). When I treated my cells with my protein of interest, Interleukin-34, there was no significant change in the expression of CD86. Unless the stimulant is changing their entire character and thus function into another lineage, the expression of this protein should remain high because it is a monocyte or of myeloid lineage.

Hi, I also have the same issue when I treat LPS 10 ng/ml to CD14+ monocytes isolated from PBMC. Want to know have you figure it out?
no, we have not pursued this property of the different clones (epitopes ?). Also because we concentrated on CD86 in the further course.
