When looking to establish Ag-specific CD8 cytotoxic T cells with autologous dendritic cells, there are numerous tumor-associated Ags to choose from. Does anybody know which Ags give the best results in terms of % of responsive CD8 T cells (e.g. gp100, tyrosinase, Melan-A... any others??) Are there great differences between these Ags in the frequency of potentially responsive CD8 T cells??

Question: When looking to establish Ag-specific CD8 cytotoxic T cells with autologous dendritic cells, there are numerous tumor-associated Ags to choose from. It depends on what the tumor is that you are targeting. My preference is for the intact antigen/sequence and using an Adenovirus vector to infect the DCs. This works for stimulation and then for targets I like to use a pox vector infected target.
If you just want to get a T-cell response to an antigen I would use tetanus or as a second choice flu. Vaccine and antigens are easy to obtain.
The more critical question is what assay to use to measure outcomes. This is especially critical for tumor antigens, which following immunization may have a T-cell clonal frequency of 1 in 50,000. I like to isolate CD8 T-cells and use an ELISPOT assay.
