I have a problem with broth dilution assay test for looking MIC of antimicrobial material. When I put my all staff(including antimicrobial materials) inside wells, then I put on my microplate on my absorbance measuring machine(it is shaking linearly every 15 minutes for 24 hours before I took absorbance measurement at 200 rpm). Candida cells started to colonize corners of the wells(shown in picture) but not in the middle of well. Also there not any turbidity in the solution which in the wells. The question is that my material has antimicrobial effect on Candida albicans or not?
