I've looking for the explanations about Ile655Val and Val655Ile in Polymorphism of HER2 breast cancer. I know Ile655Val is different with val655ile, but could you give me the explanations, which one is more harmful? Do you know of any journals?

HER2 Val655Ile allele associated with an increased risk of breast cancer, particularly in younger women.
In breast cancer patients treated with trastuzumab, the presence of Ile655Val may constitute a risk factor for cardiac toxicity.
Both of them are harmful. Second one affects the treatment choice. First one is general danger.
