For example: Culturing iPSCs in E8 and vitronectin and you only use one well on a 6-well plate, but you coated all the wells with vitronectin. After growing for several days, are those other wells still okay to use, or will the vitronectin coating degrade over time in the incubator and a fresh plate should be used?

i would not recommend to use such a plate. Better to use a freshly coated plate.
if you have this issue regularly then one option for this situation is to coat single wells of the 6W plate. Use the one that is needed and store rest of the plates at 4C (preferably 7 days - max up to 14 days). Pre-warm the coating at 37C before iPSCs are seeded.
Another alternative can be to prepare smaller aliquots of the vitronectin so that you do not have to coat extra wells. This of course depends on the amount of storage space you have.

Definitely yes. I do it constantly. It is important that vitronectin does not dry out, then it can be used. However, it depends on how long the plate is in the incubator. When vitronectin is in the incubator for up to 14 days, there is no problem with it. Sometimes even a longer period does not bother in growth hiPS cells seed on vitronectin.
