Hey everyone. I'm having gel electrophoresis problems all of a sudden and haven't been able to determine why.
The photo is a 1% TBE gel pre-stained with 10 ul gel green run at 95V for an hour. The ladders are 100bp. The rest of the samples with visible DNA should be either trnL (small) or COI amplicons (larger).
The buffer is new. Gel green is relatively new, but has been kept in a freezer box. Agarose should be fine. I've seen this ladder separate at this voltage for this amount of time.
I feel like the rig must not be working, but I've checked the connections and they seem fine. This is a replacement rig sent to me based on sending the gel rig company pics and them telling me I wasn't getting adequate current.
I'm going to try another gel rig tomorrow with the same samples.
Thoughts?
From the ladders smugging, leads me to think it is a electro chemistry thing. Make sure gel TBE, and running tbe are the same. I saw simmiler bands once when I tried to run a tbe gel in tae buffer. Also clean gel box well with di h2o.

- If the buffer new is new and your are experiencing electrophoresis problems, throw away your buffer and make again
- It does look like an electrophoresis problem as the DNA ladder has not resolved properly; Always a tell tail sign the problem lies with your agarose gel and not intrinsic to your samples
I think the most likely answer is that the buffer is at issue. Either someone made or diluted it wrong. I think the rig is probably ok, you are getting some electrophoresis and it is even across all wells.
