I want to run an SDS-PAGE of nanoparticles synthesized from plant extracts,to see the(if at all) proteins are getting adsorbed onto the nanoparticles.I have done this experiment twice but I do not get clear bands.The bands mostly diffuse to all the lanes,even the lanes with no sample shows a band.What could it be?How much likely is the presence of a plant ptotein getting adsorbed onto a nanoparticles synthesized from plant extracts?Is there a different method of sample preparations for these kinds of samples?
Hi! If I understood correctly, you have plant extracts. From these you form nanoparticles and you want to know, if all your material from the extracts has been incorporated to the particles, right? The question is, how do you form your nanoparticles? If you're crosslinking your material with eg glutaraldehyde you will probably end up with nanoparticles as well as particle building blocks (crosslinked starting material, not forming any full-blown particles). And then it all comes down to the size of your material, i.e. I presume that the mesh size of an SDS PAGE will not allow analysis of material in the nanometer size range. You could try e.g. CE or GEMMA instead to have a look at your particles. If you're interested you can have a look on our work concerning glutaraldehyde crosslinked gelatin:

Yes you got it right...the question perhaps modified.I want to check if the proteins present in the plant extract is getting adsorbed to the nanoparticles or not i.e the presence of protein as a capping/stacbilizing agent in the nanoparticle synthesized..
Hi! Ok, so I would suggest CE with UV detection instead of SDS PAGE as you can probably follow all species in your sample then.
