Hi, we are working in a very resource limited setting where the lab resources on site are very basic. We are doing serial blood sampling for drug concentrations, which includes overnight samples. Therefore, we really ave to keep blood processing to a minimum. We need plasma and PBMCs, but the PBMCs don't need to be viable. We just need to be able to isolate them to measure gene expression and drug concentrations. So my question is this: can we get away with freezing whole blood and then subsequently isolating PBMCs? We need to be able to preserve RNA. We can't really use DMSO as using a Mr frosty is probably not possible and we don't have access to liquid nitrogen. We do have a fridge, a -20C and -80C fridge.
Is this possible in any way? If not, how long can we leave the samples in the fridge for before isolating PBMCs? To what extent will isolating and freezing PBMCs that have been in the fridge for varying lengths of time (since we are doing serial sampling overnight) affect changes in gene expression?
