We collect blood by using heparin as anticoagulant instead of citrate. I want to characterize the thrombin generation potential of the PRP after our experiment. People have usually used citrated blood for thrombin assay. Would it be a problem if we use PRP from blood collected with the help of heparin for TGA assay?
Heparinized blood contains too much heparin to detect thrombin generation. TG is activated with low amounts of tissue factor and, therefore, usually not able to overcome strong anticoagulation. There are ways to neutralize heparin, but they might or will interfere with your results (and are quite expensive).
Citrate is the only recommended anticoagulant for TG measurements.

When we study thrombin generation we use citrate or oxalic acid as anti-coagulant. Never heparin because it's a strong anticoagulant drug that also in extremely low concentrations doesn't allow correct determination of thrombin generation time.
