I have my gene of interest in the pCDH cDNA Cloning and Expression Lentivectors.I understand that pCDH expression constructs are normally packaged into pseudotyped viral particles (lentiviral particles) for better transfection but can I use the same construct for simple transfection without packaging into viral particles?
Yes, absolutely, not just for pCDH, but any lentiviral vector with a constitutive promoter. However, the transfection efficiency is usually a bit lower than with a typical expression vector such as pcDNA3.1 owing to the larger size of the lentiviral vectors.

I just came across your response to Imrans question and I had a similar question recently ( for details see https://www.researchgate.net/post/Is_it_possible_to_transfect_lentiviral_transfer_plasmid_and_get_recombinant_gene_expression ). What do you exactly mean with "any lentiviral vector with a constitutive promoter" ? Do you mean a constitutive promoter in the 5' LTR as the pCDH plasmids have it or should expression after transfection also work from a constitutive internal promoter? I am currently facing problems expressing a cDNA from the pLVX plasmids (Clontech) after electroporation. The pLVX vectors use a wildtype HIV 5' LTR to drive transcription in the presence of tat from the packaging plasmid, but additionally contain an internal promoter (SFFV in my case) to drive cDNA expression. I don't see any expression after transfecting this plasmid, even though transducing it as viral particles works and also the control plasmid works after electroporation. What are your experiences with transient expression from lentiviral plasmids?
