Can we perform cell migration assays with THP1 differentiated and polarized cells?

I wanted to check the effect of my drug on the migration of M1 and M2 stages of THP1 cells. How can I do that?

Once they are polarized to M1 and M2 stages, if I trypsinize them will they still have the properties of M1 and M2 states?
