I have regularly fixed cancer cells in 70% chilled ethanol and stored them at -20 degree and processed them for PI based cell cycle analysis afterward. However, I don't know if this can be done for FACS analysis for cell surface marker. At present, I am working with ES/iPS cells and store them at -80 degree after fixing them in 4% PFA; I regularly use these cells for I-FISH ( including cell surface marker ) analysis. Therefore, I am interested to get any protocol where ES/iPS cells can be fixed and stored and then can be used for FACS even weeks after the experiment. If anyone does it (mainly for cell surface marker), please share. It will really make my life easy.
