DCPIP assay is performed for identification of hydrocarbon utilizing bacterial isolates. The color change from blue to colorless signifies the potential of the isolates to use hydrocarbon as carbon source. But, when the bacterial culture in presence of DCPIP is incubated for long time, is there any possibility that the color of the culture will turn blue again from colorless? If yes, can anyone explain me the reason behind this interesting phenomenon.
Technically speaking the colour change of DCPIP after reduction to DCPIPH2 is reversible if it can donate the electrons gained to another compound if the DCPIPH2 has not been degraded. I wouldn't be able to say if it can happen under your conditions. It can definitely not be reoxidised by oxygen
