When recive a plasmid DNA droped on the filter membrane, I crop the membrane into small pieces and put them in a 1.5 mL EP tube, then add 100 ul distilled water and keep it on 65℃ for 5-10 min (or on 4℃ for longer time) . Finally, the water can be used to transform the E. coli competant cells and grow on the resistant plate.

Step1: Cut the spot of plasmid DNA on NC membrane, transfer it into 1.5 ml MT, add 20-30ul of 10mM Tris, pH 8.5, incubate at 65 deg C in a water bath for an hour, spin at highspeed and remove the NC piece. You can use the solution as it is for further down-stream process. Check it on AGE. If you could not see follow the step 2.

Step2: Cut the NC containing the Plasmid DNA in small squares equal to the width of the well of agarose gel. Place it on the well of 1% agarose, cover it with molten agarose drop, run at 100V for few minutes, check with transilluminator upon EtBr stain. Excise the bands from the gel and do gel extraction step with any commercially available kits.

I think you will need high salt in your buffer to elute DNA

Thank you Heng Lian, Rajkumar & Anukana for the suggestions. I will try try both methods today and let you know the result. 

For elution, cut the spotted membrane and incubated at 80°C for 10 min (dry bath) in presence of 30 μl of sterile distilled water then centrifuge for 2 min at 8000 rpm
