I have experience growing dengue virus with c6/c36 cell line. However, I don’t know whether or not there is a different when it comes to growing Zika. If you have any experience or expertise in this area, I would highly appreciate if you can help me sort this out. Thank you so much for your time and expertise.

You can grow ZIKV in C6/36 and VERO cells that it will work. About the protocol it is the same. You wait until you see cytopathic effect and collect the supernatant
Thanks Julianna, we have been trying to grow ZIKV in Vero cells; however, it seems to not going well. The cells are dying very quick. Is some thing toxic to the cell from the virus? Can you send me the protocol that work well in your case?
Tara, we are not there yet. we are still trying to get the vero cells infected with ZIVK without killing the cell.
This paper grew zika in 17 different cell lines and reported the results. The efficacy was determined by PCR though rather than plaque assay. Still very helpful.

I cultured ZIKV with Vero cells and it worked well. In my case, the CPE is not that obvious and the 80% Vero cells ( including the newly grown ones) still alived after 4-days pi culture. I wonder whether you lyse the cells when harversting the viruses. This may also affect the titer.
