Can someone expliain why ADP-linked enzyme systems in tissue can't be detected by MTT assay ?

I have found that it is not possible to detect ADP-linked enzyme system in the tissue by MTT assay. Please give the explanation regarding this.

And also suggest me the suitable method for this.

The MTT assay detects reductive capacity of cells, namely NAD(P)H-dependent oxidoreductases. I'm not sure what you mean by ADP-linked enzyme systems, but if they don't involve NAD(P)H-dependent oxidoreductases then they won't be detected by the MTT assay.
