Can someone expliain why ADP-linked enzyme systems in tissue can't be detected by MTT assay ?

I have found that it is not possible to detect ADP-linked enzyme system in the tissue by MTT assay. Please give the explanation regarding this. And also suggest me the suitable method for this.
