I have isolated mitochondria from HeLa cells. I need to measure the citrate synthase activity of trhe cells. The problem is that I have no reference to compare the activity value. Any help would be greatly appreciated.
You can use Abcam citrate synthetase activity assay kit Ab119692. However, the amount of enzyme in a sample or homogenate shall depend on HeLa cells concentration i.e. their number. Use specific activity to quantify citrate synthetase activity in mitochondrial preparation.
Thank you, @Shamsher Singh Kanwar sir. I have been able to measure the CS activity in the mitochondrial extract using the procedure you mentioned. However, since I have no reference value to compare, I am unsure if my mitochondria was completely isolated and intact or not.
Prayasee, u need to purchase a commercial preparation of citrate synthetase for preparing a reference profile between CS conc or Units vs absorbance.
Use reference curve to determine CS activity in the mitochondrial preparation.
