I'd like to isolate Alternaria from plant leaves. Which media is best for rapid fungal growth as well as sporulation ? Which media give best result SDA OR PDA ?
And to induce the growth of bacteria , which media can be considered ?
Depending on the continued expertise, the medium PDA with suitable antibiotic such as chloramphenicol is very suitable for growing wide spectrum of fungi including the plant pathogenic fungi such as your fungus Alternaria spp.
Some species of the genus Alternaria like A. solani can not produce conidia on the medium PDA. Instead, you can use V8 medium to obtain the conidia of the fungus.
Hi Dr Niket Rana . Sabouraud's dextrose agar and potato-dextrose agar are commonly used media. Cultures are routinely incubated at 25° to 30° C for up to 4 weeks. Regarding bacteria, there are many suitable media for their growth. So, there are selective media for some genera and species. You must limit the genus in order to choose the selective medium. But in general, you can use Nutrient Agar, it's more suitable for wide spectrum of bacteria.
If you want to isolate the fungus Alternaria from the infected plant leaves, you must bring the infected leaves with spot disease, wash them with tap water to remove the dust, cut them to small fragments taken from the spots, sterile them with sodium hypochlorite (NaOCl) 10% for 2-3 minuets, wash them again with distilled water to remove the sterilization residue, dry them with filter paper, put these fragments on the PDA with antibiotic in the Petri dishes (5 fragment per each one) and incubate the plates under 25 C for 3-5 days.
