I am growing spheroids from HT29 colon cancer cells, and I need some sort of control cells to be compared with the HT29. Now, I should grow the control cell into spheroids and then use these new spheroids as control for metabolic rate, for instance. I found several normal cell lines, both fibroblasts and epithelial cells, but I do not know if these cell lines (CCD-33Co, FHC, CCD 841 CoN, CCD-112 CoN, CCD-18Co):
- 1: form spheroids;
- 2: are good as control cells to compare metabolic rate with a cancer cell line as HT29;
- 3: what is best between fibroblasts and epithelial cells for my application;
Hi Pilar, are you asking about non-adherent spheroid cell clusters or adherent cell masses that look like a dome?
Most low passage organ derived epithelial cells should form isolated dome like structures and not uniform monolayers like most fibroblasts.
A lot of epithelial cell lines form dome like structures at low passage (e.g. MCF7, HepG2).

Hi Steingrimur, each spheroid is a one adherent cell mass that is spheroidal, it is not a dome. So I have something between a sphere and an ellipsoid, and the entire volume of this sphere/ellipsoid is made of cells, so there are no empty spaces. I would like to know if the cell lines i mentioned before could be used to form spheroids (and not dome like structures).
HI, if fore spheroids you mean spheres in a stem cell assay yes, normal tissues contain stem cells, so they would form spheres.
About the other questions, it really depends from your scientific questions, there is no unique perfect control. If you are uncertain, you should consider all the possible control.

Hi Pilar, I also want to use these normal colon cells as control for testing toxicity in colon cancer cells. Were we able to form spheroids with any of these cell lines using scaffold free medium (using ULA microplates)?

Hi Pilar Stella Normal epithelial cells do not form smooth flat monolayers in culture. No normal epithelial cells form smooth flat monolayers in vivo like fibroblasts and endothelial cells.
True epithelial cells clump up in culture. You will have to live with that.
The price of the normal cell lines which ATCC Provides are very high. Is there any other option to find a normal coloractal cell line other than ATCC? Please suggest.

Hi Pilar Stella , I worked with FHs 74 Int (ATCC® CCL-241™) cell line and it forms spheroids on agarose coated plate.
FHs is human intestinal epithelial cell line. I used Caco-2, SW480 and HT-29 colon cancer cell line and FHs as control to check the efficiency of anticancer drug.
