I isolated T cells from mice we purchased to do an antibody titration for our flow cytometry assay. My unstimulated sample had a high CD44 expression that was down regulated when compared to the stimulated samples from the same animal. I have observed this consistently in different experiments with various mitogen? Can anybody explain or advice?

Dear Lillian,
Mitogens such as PHA, Con A reduce some surface molecules (like CD4) on daughter T cells. You can try the other stimulants for CD44 augmentation.
