I want to perform analysis on some plant samples for the detection of recombinant protein expression. For this purpose I want to use ELISA. Unfortunately I don't have a positive control. Is it possible to use my transformed Agrobacterium strain harbouring the plasmid that carries the recombinant gene?
The transformed bacterial strain should be fine if you know it is overexpressing the gene to produce the protein. If the protein of interest is intracellular, bacterial lysate should work. If the protein in soluble (i.e., released into the culture medium) and in low concenetration, you may need to use a concentrating column with the appropriate MW cut-off to increase the concentration of your protein before use. ELISAs are generally very sensitive though, so this latter step probably won't be necessary. If still in doubt, use of a good negative control such as the plant & bacterium without the insert would be valuable.
