I would like to measure the permeability of my drug using in vitro model, transwell, specifically. However, I only have the cell line, not the primary cell. Can I use the cell line to plate in the upper chamber of transwell? And I have noticed that, EGM-2 medium was used in some refrences. I also would like to know will the medium influence the intercellular junction?

there should be no problem to use EC cell line for permeability assessment in transwell system, since EC cell line also express VE-cadherin which are critical to regulate endothelial barrier function by controlling the permeability of vasculature.
