I have 500ul of sample from where I try to precipitate proteins.The sample is 2% SDS and very dilute. The conventional chl-methanol protocol suggests that I use 

1:3:1:4

volumes (sample:methanol:chloroform:water) to precipitate sample.This makes the procedure uncomfortable since the volume is very large.

My question is can I instead treat the samples as if 4 volumes of water were part of the sample, so rather than doing

500:1500:500:2000

can I instead do:

500:375:125:0? Has anyone tried that?
