We have to make a large scale cell culture of suspension cells and we were wondering if we can use regular petri dishes instead of coated cell culture dishes. Actually I have tested this before and I have obtained inhibited cell growth, however I am not sure whether it was really due to the dish. Does anybody have any information if there is an advantage for suspension cells to be grown in a cell culture dish?

I would not recommend petri dish for large scale cell culture of suspension cells. A Petri dish is a shallow transparent lidded dish that holds growth medium in which cells can be cultured. If you are going for large scale cell culture of suspension cells, petri dish will not be appropriate because it holds less volume of growth medium and there are chances of contamination. T-175 tissue culture flasks with vented caps will be most suitable for this purpose because T-175 flask can hold higher volume of growth medium and will also give you more viable cells per T-175 flask, besides reduced risk of cell contamination.

For suspension culture you need low binding special flasks, some companies supplies. Check with Fisher Scientific distributer/Nalgene etc. I have used them earlier. It has to be low binding plates. Otherwise cells can not suspend. They will attach to surface. Good luck
One consideration in dishes vs rotary culture relates to gases. Oxygen uptake may be insufficient in a still medium in a dish. And/or the pH may not be stable as cells proliferate due to insufficient exposure to CO2 in a gassed incubator. Also, concentration of medium components due to evaporation can come into play: rotary suspension vessels have larger volumes with less exposed surface.
Cultivation of clichéd cultures requires strict requirements. First, it is the nutritional value of the environment, acidity, sterility. With regard to cell subculturing, there should be no antagonism between cells if the cells are different in origin. Secondly, the number of passages of these cells and the level of differentiation should be taken into account.
