I want to ask if it is possible to store my cells 24 hours post transfection at 4 C for 4-5 days (Non-fixed) just in normal media as one of my conjugated antibody shipment is delayed and will be delivered after 2 days. I also want to ask have any one tried to fix cells with 30% methanol and store them at 4 C for 4 days and will this treatment affect antibody binding with the surface antigen?
Oof. I wouldn't. I've done immune co-culture experiments (also non-fixed cells) and found that the results even after leaving the cells overnight at 4degC weren't usable. You've got the extra variable of this being with transient transfection, too.
Unless this is really precious sample, my instinct is to just wait until the reagent arrives and do everything fresh.
I agree, I would not store cells for days at 4°C.
In my opinion, too much cells will die in this period. Moreover, proteins and other cellular structures and compounds will be degraded under this circumstances. In sum, this would/will directly distort your results.
Dear all, thanks a lot for your suggestions. I have repeated the experiment and did FACs on the fresh cells and worked well.
