I performed a fluorometric experiment with two different cell lines. I had four conditions per experiment, and four "sub-conditions" per condition, wherein I add three different chemicals (the fourth sub-condition being control). Each sub-condition was run in triplicates. The fluoreometric analysis on a spectrophotometer had different gain values and different read-outs for blank conditions. My question is, can I combine data from the same sub-condition from two different experiments?
Hi, Anand. If you normalize all the absorbance values using the control and express them as percentages, it should be fine. But if you are asking whether it is fine to combine datasets from two different cell lines into one, then it is not; even if the conditions are the same and the values are normalized.

Yes, should not combine datasets from two different cell lines. Best to repeat the experiment to get more data points if need be.
