I have not been able to find literature in which someone has used HEPES buffers in TEV protease purification. I have received advice to use sodium phosphate or Tris buffers. Any relevant literature would be welcome!
Hi Orlando, for the next time it would be good to give a little bit more information about your protein - i.e. tag type for example, resin type etc. The choice of buffer will depend on the resin type that you are going to use. In this case, I suspect the TEV protease with a his-tag. So, in case of IMAC nothing wrong with HEPES buffer, of course you can use it.
Personally, I've tried a TEV digest recently with 50 mM Tris and 50 mM Hepes overnight at 4 degrees, and both yielded identical results with my target being digested, when adding TEV to protein in a 1:2 ratio.
