Marcos Amorim Hi Marcos, thanks for your advice, I have looked at into this calculator.

It does not specify the range of the restriction enzyme. In my case, my HindIII is conventional range, but MssI is FastDigest range. The manufacture suggested any enzyme in FastDigest range are compatible in FastDigest buffer, but I am not sure if conventional HindIII can be compatible. This info is not shown on the Thermo Fisher calculator.
