I am still trying to find a protocol for preadipocyte isolation from adipose tissue. Can someone please tell me how to get PREadipocytes from adipose tissue?
Try searching for paper by Anthony Cryer but you'll need to go way back to 1980s or even late 70s. I found this one whihc might give you a place to start.
The isolation and characterization of a proposed adipocyte precursor cell type from bovine subcutaneous white adipose tissue.H A HA Plaas and A A Cryer
Fibroblast-like cells, which would be differentiated from mature adipocytes in "ceiling culture", could be isolated in primary culture. These cells precipitated, were collected from the bottom of the flask of the "ceiling culture" and further proliferated to the confluence. They finally converted to morphologically and biochemically differentiated adipocytes. In these investigations we show how to isolate fibroblast-like cells and to identify these cells as "preadipocytes"
We isolated hADSCs from human subcutaneous adipose tissue by adapting the mothod described by Gimble et al 2007 (Gimble, J.M.; Katz, A.J.; Bunnell, B.A. Adipose-derived stem cells for regenerative medicine.
Circ. Res. 2007, 100, 1249–1260.). If it helps, please find more details in our papr: Modulation of Adipogenic Conditions for Prospective Use of
hADSCs in Adipose Tissue Engineering, Int. J. Mol. Sci. 2012, 13, 15881-15900; doi:10.3390/ijms131215881.
Magnetic beads or cell sorting might help purifying the primary cell culture obtained by collagenase digestion.
