i have made niclosamide stock solution (10mM) in dmso. Further i made serial dilutions in opt- mem for my MTT assay . At concentration of 300 micromolar , i saw some yellow clouds of niclosamide coming out. Can anyone please suggest me the right solubility procedure of niclosamide. Moreover, i kept stock solution in 4 degree fridge, stock solution freeze down.
Solubility of niclosamide: DMSO (10 mM) or ethanol (25 mM).
Storage temperature.: -20°C.
Solubilise the drug in DMSO or Ethanol as Amadeu sir has said. Try also solubilising it in methanol. Then Carry out the dilutions using 1XDMEM pH 7-7.4 . See if the clouds still form.
You can dissolve 6.5 mg Niclosamide in 1ml of acetone-methanol mixture (1:1). Vortex it and it will solubilize easily. You can preserve this stock in -20 as well. Another alternative is to prepare 1 mg/ml stock in DMSO and sonicate it at 10% amplitude till the powder dissolves (2 cycles of 10 seconds each).
