I want to transfect my THP1 cells with SFTSV-NSs plasmid vector and I can not get over this ! They are floating cells so the basic Lipofectamine-based method of transfection does not work with these cells. I used Lentivirus containing my desired sequence for transfection and it is not working either. Any suggestions ?

Transfecting plasmids in THP-1 is quite challenging, not only due to the fact that they are in suspension. For example, with the Viromer RED you can expect reaching 25% of efficiency after optimization, not more. A great alternative is to transfect mRNA instead (either produced by IVT or better from an oligo supplier... it works better if you have modified mRNA).
